I have a Little Giant 4 running the return of my 225 gal reef from approximately 4.5 feet below. I would like to increase the flow at least 50%. If I add another similar pump IN-LINE with the first, will this significantly improve the gph at the return?

Ff you put it inline, youll gain a little more GPH. What you will mainly gain is more head pressure (psi)your 4.5 feet below will now actually seem like its 2.25 feet below thus gaining more GPH and PSI. Just my 2 cents. BTW i sell pumps for a living thats how i know!

I would add a separate return line. That way, you will have redundancy. In other words, if one of the pumps fails, then the other pump will maintain circulation - which just may save your tank if it happens while you are at work or away for the weekend.

If you want more flow from a centrifical pump, you should put them in parallel as gnx87 said.
